Prep SFC-MS 30
MS directed preparative SFC is fast, cost-effective, and "green". Under super-optimal conditions, a kg/kg adsorbents/day (kkd) throughput can be achieved with a preparative SFC instrument that can deliver a total flow of 400 ml/min. The reduction of solvent consumption and collection in a relatively small volume of volatile organic solvents leads to significant savings operation cost. However, the majority of the preparative SFC applications are still limited in chiral purifications, in which total flow rate determines the throughput and minimal automation is required. After witnessing the great success of MS directed preparative HPLC, it is only fitting that SFC practitioners are on a quest for an MS directed preparative SFC. The required work flow typically involves an analytical SFC-MS run on the target plate and a semi-automatic scale-up to the UV-triggered preparative SFC with a robotic work station.
Thar Instruments, makers of the TharSFC line of green chromatography systems, offers commercial MS directed preparative SFC. Its current offering, SFC-MS Prep 30, can provide users with a total flow rate up to 30 g/min and an open-bed fraction collection based on the Waters 2767 sample managing module. The system is operated on the Masslynx/Fractionlynx software platform from Waters Corporation. Using the Waters® 2767 fraction collection system in concert with MassLynx⁄FractionLynx, the system provides high throughput in a format already familiar to many chemists. This totally new preparative system features a faster, higher capacity autosampler to allow for both analytical methods development and larger preparative samples. Thus, a chemist in an open access laboratory can easily screen for a method using gradients of standard analytical flow rates of ˜5 mL⁄min and scale it up to 30 mL⁄min for prep.
